AB - Thestructuraldynamicsgoverningcollectivemotionsinoligomericmembraneproteinsplaykeyrolesinvital biomolecularprocesses atcellularmembranes.Inthisstudy,wepresentastructuralrefinementapproachthatcombinessolid-stateNMRexperimentsandmolecularsimulations toaccuratelydescribeconcertedconformationaltransitions identifying theoverallstructural,dynamical, andtopologicalstatesofoligomericmembraneproteins.Theaccuracyofthestructuralensemblesgeneratedwiththismethodisshowntoreach thestatisticalerrorlimit,andis furtherdemonstratedbycorrectlyreproducing orthogonalNMRdata. Wedemonstratetheaccuracy ofthis approachby characterisingthepentameric stateofphospholamban,akeyplayerintheregulationofcalciumuptakeinthesarcoplasmicreticulum,andbyprobingits dynamicalactivationuponphosphorylation.Ourresultsunderlinetheimportanceofusinganensembleapproachtocharacterise theconformationaltransitionsthatareoftenresponsibleforthebiologicalfunction of oligomericmembraneproteinstates.
