Polyethylenimine (PEI) has been proposed as a non-viral vector, and has been successfully used to transfer reporter genes into the central nervous system (CNS), kidneys, and lungs of adult mice. Neuropeptide Y (NPY) is a peptide expressed in the hypothalamus and is important in the regulation of body weight. Using PEI combined with stereotactic microinjection, we have successfully transferred cDNA-encoding NPY driven by the cytomegalovirus (CMV) promoter into the arcuate nucleus of adult male Wistar rats. Animals treated with NPY expressing plasmids (pNPY) gained more weight than the controls (p< 0.05), with associated increases in food intake (p<0.05) and decreased brown adipose tissue activity, measured by Guanosine Diphosphate (GDP) binding to mitochondria, (p<0.05). In a separate study, hypothalamic slices from the rats treated with pNPY/PEI showed increased NPY release (pNPY 9.7 ± 0.3 fmol/l vs. control 8.3 ± 0.5 fmol/l, p<0.05, n = 3). These results suggest that PEI is an effective vector for gene transfer into the rodent brain and can increase the protein production sufficient to result a persistent phenotypic change. This technique offers the potential of a simple and effective method to manipulate gene expression localised to specific regions of the adult rodent brain.

We have recently shown that injection of the hypothalamic peptide cocaine and amphetamine regulated transcript (CART) into discrete hypothalamic nuclei stimulates food intake. This stimulation was particularly marked in the arcuate nucleus. Here we show that twice daily intra-arcuate injection of 0.2 nmole CART peptide for 7 days was associated with a 60% higher daytime food intake, an 85% higher thermogenic response to the beta3 agonist BRL 35135, and a 60% increase in brown adipose tissue UCP-1 mRNA. In a separate study, using stereotactically targeted gene transfer, a CART transgene was delivered by using polyethylenimine to the arcuate nucleus of adult rats. Food intake was increased significantly during ad libitum feeding and following periods of food withdrawal and food restriction in CART over-expressing animals. CART over-expressing animals lost 12% more weight than controls following a 24-h fast. Brown adipose tissue uncoupling protein-1 (UCP-1) mRNA levels (collected Day 25) were 80% higher in CART over-expressing animals. Finally, by using quantitative in situ hybridization, we found that chronic cold exposure (20 days at 4oC) increased arcuate nucleus CART mRNA by 124%. Together with the orexigenic and thermogenic effects of CART, this finding suggests a role for arcuate nucleus CART in cold adaptation.

The adipocyte hormone, leptin, acts via the central nervous system to modulate glucose metabolism by skeletal muscle, but the direct effects of leptin on glucose metabolism by skeletal muscle are unclear. In this study, we have examined effects of leptin on glucose uptake by cultured L6 muscle cells assessed with the non-metabolised glucose analogue 2-deoxy-D-glucose. Reverse-transcriptase polymerase chain reaction (RT-PCR) analysis of RNA showed that L6 muscle cells express a short isoform of the leptin receptor (ObRa), but not the long isoform (ObRb). In the absence of added insulin, incubation of L6 muscle cells with murine leptin (10-11- 10-8 M) for 10 min and 1 h increased glucose uptake by 15%-23%. This effect of leptin was lost by 4 h. Leptin (10-10 - 10-9 M) initially (after 10 min) suppressed insulin-stimulated glucose uptake by 14-16%, but had no effect in the longer term. Leptin-stimulated glucose uptake was inhibited by the phosphatidylinositol 3-kinase (PI3K) inhibitor wortmannin, but not by the janus kinase-2 (JAK-2) inhibitor tyrphostin AG490. The results suggest that leptin can act directly on L6 muscle cells via a short leptin receptor isoform to acutely stimulate basal (but not insulin-stimulated) glucose uptake via a PI3K-dependent pathway.
