NOD1 variation, immunoglobulin E and asthma
, Human Molecular Genetics, Vol: 14, Pages: 935-941, ISSN: 1460-2083Asthma is a familial inflammatory disease of the airways of the lung. Microbial exposures in childhood protectagainst asthma through unknown mechanisms. The innate immune system is able to identify microbial componentsthrough a variety of pattern-recognition receptors (PRRs). NOD1 is an intracellular PRR that initiatesinflammation in response to bacterial diaminopimelic acid (iE-DAP). The NOD1 gene is on chromosome 7p14, ina region that has been genetically linked to asthma. We carried out a systematic search for polymorphism in thegene. We found an insertion–deletion polymorphism (ND1 1 32656) near the beginning of intron IX thataccounted for 7% of the variation in IgE in two panels of families (P < 0.0005 in each). Allele*2 (the insertion)was associated with high IgE levels. The same allele was strongly associated with asthma in an independentstudy of 600 asthmatic children and 1194 super-normal controls [odds ratio (OR) 6.3; 95% confidence interval(CI) 1.4–28.3, dominant model]. Differential binding of the two ND1 1 32656 alleles was observed to a proteinfrom nuclei of the Calu 3 epithelial cell line. In an accompanying study, the deletion allele (ND1 1 32656*1)was found to be associated with inflammatory bowel disease. The results indicate that intracellular recognitionof specific bacterial products affects the presence of childhood asthma.

Wetting of bioactive glass surfaces by poly(α-hydroxyacid) melts: Interaction between Bioglass® and biodegradable polymers
, E Polymers, ISSN: 1618-7229The interfacial characteristics between bioactive glass (45S5 Bioglass®) surfaces and poly(α-hydroxyacid) melts have been assessed by direct wetting measurements. In particular, the wettability of Bioglass® powder by poly(D,L-lactide) (PDLLA) and poly(D,L-lactide-co-glycolide) (PLGA) was assessed by imbibition measurements. Additionally, the equilibrium contact angles of PDLLA and PLGA melts on a sintered Bioglass® surface were measured. The surface energy of the bioactive glass and the polymers was determined from contact angles measured using various test liquids on PDLLA, PLGA and Bioglass® solid substrates. There are sufficient adhesive interactions between the polymers and Bioglass®. A simple heat treatment of the bioactive glass in an inert gas atmosphere leads to an improved wetting behaviour, indicating increased adhesive interactions. Scanning electron micrographs of the polymer + Bioglass® composites formed by polymer penetration into the powder bed show the formation of a 'good quality' interface.

On the origins of extreme mycorrhizal specificity in the Monotropoideae (Ericaceae): performance trade-offs during seed germination and seedling development.
, Molecular Ecology, Vol: 14, Pages: 1549-1560, ISSN: 0962-1083Fungal-induced seed germination is a phenomenon characteristic of mycorrhizal plants that produce dust-like seeds with only minimal nutritional reserves. In such systems, fungi trigger germination and/or subsidize development. We studied mycorrhizal germination in relation to mycorrhizal specificity in the Monotropoideae, a lineage of dust-seeded non-photosynthetic plants that are dependent upon ectomycorrhizal fungi of forest trees. A total of 1695 seed packets, each containing two to five compartments with seeds from different sources, were buried for up to 2 years near known ectomycorrhizal fungi in six different native forest locations. Upon harvest, seedlings were analysed by cultivation-independent molecular methods to identify their mycorrhizal fungi. We report that (i) germination is only induced by the same fungus that associates with mature plants or by closely related congeners; (ii) seedlings associated with the latter fungi develop less than those associated with maternal fungal species in most settings; and (iii) exceptions to this pattern occur in allopatric settings, where novel plant-fungal associations can result in the greatest seedling development. We interpret these results as evidence of performance trade-offs between breadth of host range and rate of development. We propose that in conjunction with host-derived germination cues, performance trade-offs can explain the extreme mycorrhizal specificity observed at maturity. The allopatric exceptions support the idea that performance trade-offs may be based on a coevolutionary arms race and that host range can be broadened most readily when naive fungal hosts are encountered in novel settings.

Characterisation of 'wet' polymer surfaces for tissue engineering applications: Are flat surfaces a suitable model for complex structures?
, E Polymers, ISSN: 1618-7229A study of the morphorlogy and surface chemistry of poly(D,L-lactide) (PDLLA) substrates, fabricated into two different structures, was presented. A 3D, porous, foam-like scaffold prepared by the thermally induced phase separation (TIPS) method and flat polymer thin films made by solvent casting was studied. A method to assess the wettability of wet pore wall surfaces inside highly porous 3D structures, which was based on the maximum bubble point test, was developed and tested. The results show that PDLLA contains acidic functional groups and is hydrophobic.
