Affinity of various benzodiazepine site ligands in mice with a point mutation in the GABA(A) receptor gamma2 subunit.
, Biochem Pharmacol, Vol: 68, Pages: 1621-1629, ISSN: 0006-2952The benzodiazepine binding site of GABA(A) receptors is located at the interface of the alpha and gamma subunits. Certain point mutations in these subunits have been demonstrated to dramatically reduce the affinity of benzodiazepine binding site ligands for these receptors. Recently, mice were generated with a phenylalanine (F) to isoleucine (I) substitution at position 77 in the gamma2 subunit of GABA(A) receptors. Here we tested the potency of 24 benzodiazepine binding site ligands from 16 different structural classes for inhibition of [(3)H]flunitrazepam binding to brain membranes of these gamma2F77I mice. Results indicate that the potency of the classical 1,4-benzodiazepines, of the 1,4-thienodiazepine clotiazepam, the 1,5-benzodiazepine clobazam, or the pyrazoloquinoline CGS 9896 is only 2-7-fold reduced by this gamma2F77I point mutation. The potency of the imidazopyrimidines Ru 32698, Ru 33203, and Ru 33356, of the imidazoquinoline Ru 31719, or the pyrazolopyridine CGS 20625 is reduced 10-20-fold, whereas the potency of some imidazobenzodiazepines, beta-carbolines, cyclopyrrolones, imidazopyridines, triazolopyridazines, or quinolines is 100-1000-fold reduced. Interestingly, the extent of potency reduction induced by the gamma2F77I point mutation varied within the structural classes of compounds. Results support and significantly extend previous observations indicating that the residue gamma2F77 is important for high affinity binding of some, but not all benzodiazepine site ligands.

VEMAP Phase 2 bioclimatic database. I. Gridded historical (20th century) climate for modeling ecosystem dynamics across the conterminous USA
, Climate Research, Vol: 27, Pages: 151-170, ISSN: 0936-577XAnalysis and simulation of biospheric responses to historical forcing require surface climate data that capture those aspects of climate that control ecological processes, including key spatial gradients and modes of temporal variability. We developed a multivariate, gridded historical climate dataset for the conterminous USA as a common input database for the Vegetation/Ecosystem Modeling and Analysis Project (VEMAP), a biogeochemical and dynamic vegetation model intercomparison. The dataset covers the period 1895-1993 on a 0.5° latitude/longitude grid. Climate is represented at both monthly and daily timesteps. Variables are: precipitation, mininimum and maximum temperature, total incident solar radiation, daylight-period irradiance, vapor pressure, and daylight-period relative humidity. The dataset was derived from US Historical Climate Network (HCN), cooperative network, and snowpack telemetry (SNOTEL) monthly precipitation and mean minimum and maximum temperature station data. We employed techniques that rely on geostatistical and physical relationships to create the temporally and spatially complete dataset. We developed a local kriging prediction model to infill discontinuous and limited-length station records based on spatial autocorrelation structure of climate anomalies. A spatial interpolation model (PRISM) that accounts for physiographic controls was used to grid the infilled monthly station data. We implemented a stochastic weather generator (modified WGEN) to disaggregate the gridded monthly series to dailies. Radiation and humidity variables were estimated from the dailies using a physically-based empirical surface climate model (MTCLIM3). Derived datasets include a 100 yr model spin-up climate and a historical Palmer Drought Severity Index (PDSI) dataset. Changing partners in the dark: isotopic and molecular evidence of ectomycorrhizal liaisons between forest orchids and trees.
, Proceedings of the Royal Society B, Vol: 271, Pages: 1799-1806, ISSN: 0962-8452In the mycorrhizal symbiosis, plants exchange photosynthates for mineral nutrients acquired by fungi from the soil. This mutualistic arrangement has been subverted by hundreds of mycorrhizal plant species that lack the ability to photosynthesize. The most numerous examples of this behaviour are found in the largest plant family, the Orchidaceae. Although these non-photosynthetic orchid species are known to be highly specialized exploiters of the ectomycorrhizal symbiosis, photosynthetic orchids are thought to use free-living saprophytic, or pathogenic, fungal lineages. However, we present evidence that putatively photosynthetic orchids from five species which grow in the understorey of forests: (i) form mycorrhizas with ectomycorrhizal fungi of forest trees; and (ii) have stable isotope signatures indicating distinctive pathways for nitrogen and carbon acquisition approaching those of non-photosynthetic orchids that associate with ectomycorrhizal fungi of forest trees. These findings represent a major shift in our understanding of both orchid ecology and evolution because they explain how orchids can thrive in low-irradiance niches and they show that a shift to exploiting ectomycorrhizal fungi precedes viable losses of photosynthetic ability in orchid lineages.
