STRUCTURAL BASIS OF LECTIN-CARBOHYDRATE RECOGNITION
, Annual Review of Biochemistry, Vol: 65, Pages: 441-473, ISSN: 0066-4154<jats:p> Lectins are responsible for cell surface sugar recognition in bacteria, animals, and plants. Examples include bacterial toxins; animal receptors that mediate cellcell interactions, uptake of glycoconjugates, and pathogen neutralization; and plant toxins and mitogens. The structural basis for selective sugar recognition by members of all of these groups has been investigated by x-ray crystallography. Mechanisms for sugar recognition have evolved independently in diverse protein structural frameworks, but share some key features. Relatively low affinity binding sites for monosaccharides are formed at shallow indentations on protein surfaces. Selectivity is achieved through a combination of hydrogen bonding to the sugar hydroxyl groups with van der Waals packing, often including packing of a hydrophobic sugar face against aromatic amino acid side chains. Higher selectivity of binding is achieved by extending binding sites through additional direct and water-mediated contacts between oligosaccharides and the protein surface. Dramatically increased affinity for oligosaccharides results from clustering of simple binding sites in oligomers of the lectin polypeptides. The geometry of such oligomers helps to establish the ability of the lectins to distinguish surface arrays of polysaccharides in some instances and to crosslink glycoconjugates in others. </jats:p>

It crystallizes in the monoclinic space group C2/c with a= 18.139-(8) Å, b = 12.694(5) Å, c - 17.694(8) Å, β = 107.5(6)°, V = 3885(6) Å<sup>3</sup>, and Z = 4. The cation [Mn<inf>2III Iv</inf>O<inf>2</inf>(bisimMe<inf>2</inf>en)<inf>2</inf>]<sup>3+</sup> contains a di-manganese di-μ-oxo unit. The Mn-Mn axis is a C<inf>2</inf> axis. The Mn-imidazole distances for Mn<sup>III</sup> or Mn<sup>IV</sup> are distinct: respectively, 2.208(9) and 2.004(8) Å. The ground state is a |S<inf>A</inf>=2,S<inf>B</inf>=3/2,S=1/2〉 state separated by 420 cm<sup>-1</sup> from the S = 3/2 state. The EPR spectrum has been simulated with |A<inf>1tA</inf>| = 160.10<sup>-4</sup>cm<sup>-1</sup>, |A<inf>1Z</inf>,.-| = 136.10<sup>-4</sup>Cm<sup>-1</sup>, \A<inf>2tA</inf>| = 72.10<sup>-4</sup> cm<sup>-1</sup>, |A<inf>2z</inf>|= 71.10<sup>-4</sup> cmn<sup>-1</sup>, g<inf>-ty</inf> = 1.997. g-= 1.993. The ESEEM spectrum is reported. The <sup>2</sup>H nuclei corresponding to the exchangeable hydrogen atoms of imidazole groups in D<inf>2</inf>O have been detected at 2.4 MHz at 3480 G.
