Characterisation of the low-fluorescent (LF1) mutant of Scenedesmus by EPR
, Bba Bioenergetics, Vol: 932, Pages: 171-176, ISSN: 0005-2728An EPR study of the low fluorescent mutant, LF1, of the green alga, Scenedesmus obliquus, has been performed and the following results were obtained. (1) Stable photoinduced charge separation occurs at low temperature forming the characteristic EPR signals from Q<sup>-</sup><inf>A</inf>Fe<sup>2+</sup>, the primary semiquinone-iron complex, indicating no major structural modification of the electron acceptor complex in the LF1 mutant. (2) The source of the electron in this reaction at 200 K is a component, probably a chlorophyll molecule, which gives rise to an EPR signal at g ≈ 2.0026 when photooxidized; in the wild type the S<inf>2</inf> manganese multiline EPR signal is generated under these conditions. (3) Contrary to a previous report (Biochim. Biophys. Acta 682 (1982) 106-114), Signal II is present in the dark in LF1; however, it exhibits different microwave power saturation characteristics from those of Signal II in the wild type. These results show that the LF1 mutant has a functional reaction centre which lacks the redox active manganese of the O<inf>2</inf>-evolving enzyme. In addition, as reported previously, cytochrome b-559 is in the low-potential, oxidized form in LF1, while it is largely in the high-potential, reduced form in the wild-type. This effect is thought to be only indirectly related to the 34 kDa to the 36 kDa change in D<inf>1</inf> polypeptide size, which is assumed to be responsible for the lack of Mn binding in the LF1 mutant. © 1988.

Simulation of regional soil moisture deficits on a European scale
, Norsk Geografisk Tidsskrift, Vol: 42, Pages: 149-151, ISSN: 0029-1951A simple deterministic simulation model for landscape-scale soil water deficits has been developed. Standard weather station data are used to estimate ‘daily’ precipitation and actual (regional) evapo- ranspiration (AET). The balance is added to soil moisture, which is accounted by a simple bucket nodel. AET is the daily integral of the lesser of supply and demand: Supply is governed by current oil moisture, demand by estimated net radiation and temperature. The model estimates geographical atterns of soil water deficit at a scale suitable for comparison with the distributions of plant species md vegetation types. The model provides a way of quantifying soil moisture stress in biogeographical esearch. © 1988 Taylor & Francis Group, LLC.

Structural and functional basis for GABAA receptor heterogeneity
, Nature, Vol: 335, Pages: 76-79, ISSN: 0028-0836When gamma-aminobutyric acid (GABA), the major inhibitory neurotransmitter in vertebrate brain, binds to its receptor it activates a chloride channel. Neurotransmitter action at the GABAA receptor is potentiated by both benzodiazepines and barbiturates which are therapeutically useful drugs (reviewed in ref. 1). There is strong evidence that this receptor is heterogeneous. We have previously isolated complementary DNAs encoding an alpha- and a beta-subunit and shown that both are needed for expression of a functional GABAA receptor. We have now isolated cDNAs encoding two additional GABAA receptor alpha-subunits, confirming the heterogeneous nature of the receptor/chloride channel complex and demonstrating a molecular basis for it. These alpha-subunits are differentially expressed within the CNS and produce, when expressed with the beta-subunit in Xenopus oocytes, receptor subtypes which can be distinguished by their apparent sensitivity to GABA. Highly homologous receptor subtypes which differ functionally seem to be a common feature of brain receptors.

Distinct GABAA receptor alpha subunit mRNAs show differential patterns of expression in bovine brain
, Neuron, Vol: 1, Pages: 937-947, ISSN: 0896-6273Specific oligonucleotide probes have been used to visualize the regional and cellular distribution of the mRNAs encoding three structurally distinct GABAA receptor alpha subunits in bovine brain. In situ hybridization analysis showed that these transcripts differ in distribution and in relative abundance. In frontal cortex the alpha 1 and alpha 2 transcripts are most abundant in layers II-IV, whereas the alpha 3 mRNA is most abundant in layers V and VI. In the hippocampal complex, the alpha transcripts are differentially distributed in the entorhinal cortex and subiculum. The alpha 2 transcript is enriched in the dentate gyrus and CA4/CA3 regions of the hippocampus. In the cerebellum, essentially only the alpha 1 transcript is detectable in granule cells, Purkinje cells, and stellate/basket cells. These results suggest that the different alpha subunits represent components of distinct GABAA receptor subtypes.

Engineering bacterial toxin for the development of new vaccine against pertussis.

The cellular pertussis vaccine introduced in the forties is highly effective and is widely used, but its reactogenicity has led to public concern regarding its safety. The attempts to reduce the side effects associated with pertussis immunization have led to the preparation of acellular B. pertussis products: one composed of detoxified pertussis toxin (PT) and filamentous haemagglutinin (FHA), another one composed only of detoxified PT and a third vaccine composed of detoxified PT, FHA and serotypes 1, 3 of fimbriae. In our laboratories we have approached the study of pertussis toxin, the molecule present in all the proposed acellular pertussis vaccines and one of the main virulence factors of B. Pertussis, with the aim of producing new acellular pertussis vaccines by using recombinant DNA techniques.
