Pollen percentages, tree abundances and the Fagerlind effect
, Journal of Quaternary Science, Vol: 1, Pages: 35-43, ISSN: 0267-8179Pollen diagrams traditionally are read as if pollen percentages were linearly related to relative tree abundances, although the slopes and intercepts of these relationships are accepted to differ among taxa. Corresponding map patterns of modern pollen and tree percentages support this assumption of linearity, which also underlies the use of linear regression on percentage data for pollen‐tree calibration. Fagerlind, however, showed that the theoretical relationship need not be linear and may be confounded by interdependencies among taxa. Regressions and scatter plots of modern pollen and tree percentages are here compared with results of extended R‐value (ERV) models, which correct for the ‘Fagerlind effect’. Three data sets from Wisconsin and Michigan, USA illustrate that regression coefficients provide a first approximation to their ERV equivalents, but scatter plots derived from the ERV analyses show reduced scatter about linearised relationships. The pharmacology of simple molecules.
, Arch Toxicol Suppl, Vol: 9, Pages: 27-37, ISSN: 0171-9750The biological effects of simple molecules have traditionally been ascribed to their actions on the lipid portions of biological membranes. However, at the low concentrations of these molecules which induce general anaesthesia or have acute toxic effects in animals, changes in lipid bilayer properties are so small that they are unlikely to be relevant biologically. On the other hand, these molecules do inhibit the activity of a pure lipid-free protein, with ED50 concentrations which are essentially identical to the biological ED50 and LD50 concentrations. Moreover, the well-known but puzzling cutoffs in potencies in homologous series of compounds are also found with this enzyme. The accumulating evidence now suggests that the pharmacological effects of low concentrations of relatively inert agents are best explained in terms of their direct binding to amphiphilic pockets of circumscribed dimensions on proteins.

Characterization of peptides cleaved by plasmin from the C-terminal polymerization domain of human fibrinogen.
, J Biol Chem, Vol: 260, Pages: 13095-13101, ISSN: 0021-9258The C-terminal region of the fibrinogen gamma chain is known to participate in several functional interactions including fibrin polymerization. This part of the molecule is retained on the gamma chain of fragment D (FgD) when fibrinogen is digested by plasmin in the presence of calcium to produce the fragment D-fragment E (FgD X FgE) complex but is lost if FgD is prepared in the absence of calcium. In an attempt to characterize the C-terminal polymerization domain we have used three techniques to examine this further degradation of FgD following the addition of EDTA and plasmin. Analysis of the digestion by sodium dodecyl sulfate-polyacrylamide gel electrophoresis showed a progressive cleavage of the gamma chain to two small remnants. The polymerization-inhibitory activity of the whole digest was studied using acid-solubilized fibrin. A progressive loss of inhibitory activity was associated with gamma chain shortening, reaching greater than a 120-fold reduction at the end of digestion. The cleavage of peptides was followed by reverse-phase high performance liquid chromatography and the release of a characteristic peptide triplet was associated with gamma chain cleavage. Manual sequencing, amino acid analysis, and fast atom bombardment mass spectrometry established the three peptides as gamma 303-356, 357-373, and 374-405. These peptides have sequences in common with those peptides recently reported by other investigators to be potent polymerization inhibitors. However, when a mixture of the three peptides was added in a 200-fold molar excess to polymerizing fibrin, no inhibitory activity could be demonstrated. It is concluded that the C-terminal polymerization domain of fibrinogen may be an extended region which includes the sequence gamma 303-405, when this is contiguous with the remainder of the gamma chain.
