AB - NMR spectroscopy is the most comprehensive analytical tool for chemical structure elucidation and verification. This article aims at introducing and explaining the basics of the most useful NMR pulse sequences for structural elucidation of small organic molecules such as metabolites, drugs and natural products. Step by step we introduce the experiments that are needed to determine the backbone structure and stereochemistry, terminating with a brief description of some of the latest developments in pulse sequences for improving spectral resolution and acquisition. The described experiments are available in most modern NMR spectrometers, from high-resolution systems to benchtop systems.
