A comprehensive dynamic model to describe monoclonal antibody (MAb)-producing mammalian cell cultures is presented featuring: (i) cell growth kinetics, (ii) cell metabolism, (iii) MAb synthesis and secretion and (iv) MAb glycosylation. Its simulation results are in good agreement with experimental data for macroscopic culture variables and qualitative observations for glycosylation. The combined model allows us to examine in silico and a priori the impact of feeding schedules on the extent of MAb glycosylation and productivity. Optimisation results for different feeding strategies and their effect on glycosylation patterns are also presented, based on which product quality monitoring strategies can be developed.

This paper presents an overview of meoretical and algorithmic developments in the area of parametric programming and control. The application of parametric programming for the explicit solution of model based control problems in the presence of constraints is discussed in detail. These concepts are illustrated with a biomedical system control problem concerning the regulation of blood glucose concentration for people with type 1 diabetes.

In this work, a parametric programming based approach is proposed to the hierarchical plant selection problem. Based on recently proposed multi-parametric programming algorithms (Dua et al., 2002; Dua and Pistikopoulos, 2000), multi-level programming problems can be solved without disassembling the multi-level nature of them, since the rational reaction sets can be computed in advance and thereby the leader can choose the global optimum. Moreover, since the rational reaction set is computed through the Basic Sensitivity Theorem (Fiacco, 1983), the behaviour of the follower will be given by an affine function of the leader's variables. Consequently, in the upper level no extra numerical complexity is introduced. The advantage of this technique is that the parametric nature of the leader's problem is preserved and allows for possible comparisons between global optima. The underlying mathematical theory and a illustrating example are provided.

Fuel cell power systems have exceptional potential and applications for both stationary and mobile applications due to their outstanding energy efficiency and environmental characteristics. A discussion covers the development of a parametric controller for a solid oxide fuel cell (SOFC) system; parametric control of the SOFC system temperature; control action function for each of the subregions; basis for the controller design problem; model identification and reduction methodologies; and temperature control via suitable manipulation of the fuel and oxidant flowrates, and the use of the produced voltage signal. This is an abstract of a paper presented at the 2006 AIChE Annual Meeting (San Francisco, CA 11/12-17/2006).

Dynamic oil and gas production systems simulation and optimization is a research trend with a potential to meet the challenges faced by the international oil and gas industry, as has been already demonstrated in a wide variety of publications in the open literature. The complex two-phase flow in reservoirs and production wells governs fuel transport, but is mostly handled by algebraic approximations in modern optimization applications; the true reservoir state variable profiles (initial/boundary conditions) are not known. Integrated modeling and optimization of oil and gas production systems treats oil reservoirs, wells and surface facilities as a single (yet multiscale) system, focusing on computing accurate reservoir and well state variable profiles, useful for optimization. This paper discusses a strategy for interfacing reservoir simulation (ECLIPSE®) with equation-oriented process optimization (gPROMS®) and presents a relevant application.

In this paper model based control for two drug delivery systems is presented: (i) simultaneous control of mean arterial pressure (MAP), cardiac output (CO) and hypnosis for patients undergoing surgery under anesthesia and (ii) blood glucose control for type 1 diabetics. A compartmental model is presented for delivery of drugs under anesthesia. Advanced model based controllers are then derived for the simultaneous regulation of MAP, CO and bispectral index (BIS). For type 1 diabetics, optimal insulin delivery rate is obtained as an explicit function of the state of the patient by using parametric programming. This reduces the implementation of the model based controller to simple function evaluations.
