Apolipoprotein E (APOE) plays an important role in the multifactorial etiology of both cardiovascular disease and Alzheimer's disease. Polymerase chain reaction (PCR) was used to investigate the APOE gene polymorphism in 335 unrelated Greek Cypriots living on the island of Cyprus. For the most common APOE genotypes, the Greek Cypriots followed the general Caucasian European pattern of having higher genotypic frequencies of E3/3, followed by E3/4, and then E2/3. Among the European populations compared, Greek Cypriots exhibited the lowest relative frequency of the E3/4 genotype (12.83%). Also, the relative frequencies of the E2 and E4 alleles in Greek Cypriots were among the lowest around the world (5.4% and 7.0%, respectively). This was also demonstrated by using the complete and the average clustering methods of analysis where the APOE allele relative frequencies in Greek Cypriots were compared to 46 other populations. The Greek Cypriot population in these analyses clustered with populations mainly from south Europe and Japan which have low E2 and E4 allele frequencies. The Greek Cypriot population will be studied further for elucidating the effect(s) and the role of APOE in cardiovascular disease and the APOE4 allele as a possible metabolic factor affecting the rate of expression of both Alzheimer's disease and vascular dementia.

Bispectral analysis suggests that EMG is highly non-Gaussian at force level (FL) = 10% of maximum voluntary contraction; the value of the test statistic for Gaussianity decreases with increasing FL up to 70%, and then slightly increases at FL = 100%. Furthermore the magnitude of the bispectrum increases with force.

Several lines of evidence suggest that many cases of Alzheimer's disease (AD) may be due to genetic factors. We used subtraction hybridization to isolate genes that were differentially expressed in AD compared to control brains. Directionally cloned cDNA libraries from AD and control patients' cerebral temporal cortices were used for the production of sense and antisense cDNA using the polymerase chain reaction (PCR). A 30- to 40-fold excess of sense cDNA from Alzheimer's disease brain was photobiotinylated and hybridized to (32)P-labeled antisense cDNA from control brain. The hybridized and unhybridized "driver" DNA was removed by streptavidin binding and phenol extraction. The subtracted antisense cDNA sequences were PCR amplified and cloned into lambda-GEM-4 producing a subtracted cDNA library. This subtracted cDNA library was rescreened using duplicate differential colony hybridization to select specific subtracted clones. An 850-bp cDNA that was overexpressed in the AD compared to the control library was isolated and sequenced. It had >95% homology to cytochrome oxidase subunit 3. Overexpression of this gene in AD brains was confirmed using Northern blots. Since cytochrome oxidase is important for neuronal function, these findings suggest a possible role for this gene in the pathogenesis of AD.

The diagnostic usefulness of artificial neural networks (ANNs) is explored by means of an integrated system for medical diagnosis. The models were developed by using the unsupervised self organizing feature maps algorithm. Clinical and laboratory data for training and testing the ANNs was collected from 71 subjects by applying examination protocols that were developed by experts in the appropriate fields. The diagnostic yield obtained by the examined models was in the region of 80 to 90%.

Neural networks, genetic algorithms, and the K-means clustering algorithm have been used in this study for the classification of quantitative electromyographic data. The results that have been independently obtained by the above methods are presented and the relative advantages and disadvantages are discussed. The comparative study of these methods guidance in choosing the most appropriate method for the classification of electromyographic data. It is also suggested that the problem of medical diagnosis can be handled better by combining more than one method.

The prevalence of multiple sclerosis (MS) was studied in the Greek-speaking population in the southwestern district of Paphos and the eastern Famagusta area and in the Troodos mountains in Cyprus. Excluding refugees from the northern part of the island, the MS prevalence was 44.5 per 100,000. This is a similar prevalence to that found in Sicily and shows that, like Sicily, Greek-speaking Cypriots have a medium to high and not a low MS prevalence. There was no significant difference in MS prevalence between the warm Paphos and Famagusta areas and the much colder mountainous areas. In one village, Liopetri, there were five families with definite MS (CDMS), and one patient with clinically probable MS (CPMS). Excluding refugees, the prevalence for CDMS and CPMS in this village was 240 per 100,000. All six families are probably related.
