To explore in vitro anticancer potential of Aerva lanata L. (flowering aerial part). The study was performed with 5 different human cell lines for the study of lung, leukaemia, prostate, colon and cervix cancer by using Sulphorhodamine B (SRB) assay. There were three doses of 10, 30 and 100 microg mL(-1) of each Aerva lanata L. Chloroform fraction (ALCF) and Aerva lanata L. Ethyl Acetate Fraction (ALEAF) used in this study. ALCF showed significant % inhibitory effect for leukaemia, lung and colon cancer at maximum concentration of 100 microg mL(-1) as compared to standard drug mitomycin. On the other hand ALEAF showed the significant % inhibitory effect for lung and cervix cancer at maximum concentration of 100 microg mL(-1) as compared to standard drug 5-fluoro Uracil (5-FU). From the above studies it is concluded that, the ethyl acetate fraction and chloroform fraction of Aerva lanata L. provide enough experimental evidence for anticancer activity and these fractions could be useful in medical care.

BACKGROUND: The objectives of the study were to compare the performance of line probe assay (GenoType MTBDRplus) with solid culture method for an early diagnosis of multidrug resistant tuberculosis (MDR-TB), and to study the mutation patterns associated with rpoB, katG and inhA genes at a tertiary care centre in north India. METHODS: In this cross-sectional study, 269 previously treated sputum-smear acid-fast bacilli (AFB) positive MDR-TB suspects were enrolled from January to September 2012 at the All India Institute of Medical Sciences hospital, New Delhi. Line probe assay (LPA) was performed directly on the sputum specimens and the results were compared with that of conventional drug susceptibility testing (DST) on solid media [Lowenstein Jensen (LJ) method]. RESULTS: DST results by LPA and LJ methods were compared in 242 MDR-TB suspects. The LPA detected rifampicin (RIF) resistance in 70 of 71 cases, isoniazid (INH) resistance in 86 of 93 cases, and MDR-TB in 66 of 68 cases as compared to the conventional method. Overall (rifampicin, isoniazid and MDR-TB) concordance of the LPA with the conventional DST was 96%. Sensitivity and specificity were 98% and 99% respectively for detection of RIF resistance; 92% and 99% respectively for detection of INH resistance; 97% and 100% respectively for detection of MDR-TB. Frequencies of katG gene, inhA gene and combined katG and inhA gene mutations conferring all INH resistance were 72/87 (83%), 10/87 (11%) and 5/87 (6%) respectively. The turnaround time of the LPA test was 48 hours. CONCLUSION: The LPA test provides an early diagnosis of monoresistance to isoniazid and rifampicin and is highly sensitive and specific for an early diagnosis of MDR-TB. Based on these findings, it is concluded that the LPA test can be useful in early diagnosis of drug resistant TB in high TB burden countries.

Background & AimsOutcome prediction is uniquely different in hepatocellular carcinoma (HCC) as the progressive functional impairment of the liver impacts patient survival independently of tumour stage. As chronic inflammation is associated with the pathogenesis of HCC, we explored the prognostic impact of a panel of inflammatory based scores, including the modified Glasgow Prognostic Score (mGPS), neutrophil to lymphocyte ratio (NLR) and platelet to lymphocyte ratio (PLR), in independent cohorts.MethodsInflammatory markers, Barcelona Clinic Liver Cancer (BCLC) and Cancer of the Liver Italian Program (CLIP) scores were studied in a training set of 112 patients with predominantly unresectable HCC (75%). Independent predictors of survival identified in multivariate analysis were validated in an independent cohort of 466 patients with an overall lower tumour burden (BCLC-A, 56%).ResultsIn both training and validation sets, mGPS and CLIP scores emerged as independent predictors of overall survival. The predictive accuracy of the combined mGPS and CLIP score (c score 0.7, 95% CI 0.6–0.8) appeared superior to that of the CLIP score alone (c score 0.6, 95% CI 0.5–0.7).ConclusionsSystemic inflammation as measured by the mGPS, independently predicts overall survival in HCC. We have validated a novel, easy to use inflammatory score that can be used to stratify individuals. These data enable formulation of a new prognostic system, the inflammation based index in HCC (IBI). Further validation of the IBI considering treatment allocation and survival is warranted in an independent patient cohort.

Recent large--scale association studies, both of genome-wide and candidate gene design, have revealed several single-nucleotide polymorphisms (SNPs) which are significantly associated with risk of developing breast cancer. As both breast and endometrial cancers are considered to be hormonally driven and share multiple risk factors, we investigated whether breast cancer risk alleles are also associated with endometrial cancer risk. We genotyped nine breast cancer risk SNPs in up to 4188 endometrial cases and 11,928 controls, from between three and seven Caucasian populations. None of the tested SNPs showed significant evidence of association with risk of endometrial cancer.

BackgroundPlatinum agents have shown demonstrable activity in the treatment of patients with platinum resistant, recurrent ovarian cancer when delivered in a "dose-dense" fashion. However, the development of thrombocytopenia limits the weekly administration of carboplatin to no greater than AUC 2. Paclitaxel has a well-described platelet sparing effect however its use to explicitly provide thromboprotection in the context of dose dense carboplatin has not been explored.MethodsWe treated seven patients with platinum resistant ovarian cancer who had previously received paclitaxel or who had developed significant peripheral neuropathy precluding the use of further full dose weekly paclitaxel.ResultsWe were able to deliver carboplatin AUC 3 and paclitaxel 20 mg/m2 with no thrombocytopenia or worsening of neuropathic side-effects, and with good activity.ConclusionsWe conclude that this regimen may be feasible and active, and could be formally developed as a "platinum-focussed dose-dense scaffold" into which targeted therapies that reverse platinum resistance can be incorporated, and merits further evaluation.

This study reported the purification of acetone extract of Pyrus pashia Buch.-Ham. ex D. Don stem bark. The powdered drug was defatted with petroleum ether and then extracted with acetone. The orange brown acetone extract had no odour, showed the presence of desired phytochemicals i.e. steroids, tannins, terpenes etc. The acetone extract was further subjected to chromatographic purification in separating funnel and one pure compound has been successfully isolated from acetone fraction which was confirmed on the basis of TLC, paper chromatography. UV spectroscopic studies were also performed on isolated compound. This method of purification involving separating funnel was employed for the first time and found to be simple, cost effective and highly efficient for purification of acetone extract of Pyrus pashia Buch.-Ham. ex D. Don stem bark.
