Possible Regulatory Roles of Promoter G-Quadruplexes in Cardiac Function-Related Genes - Human TnIc as a Model
, PLOS One, Vol: 8, ISSN: 1932-6203G-quadruplexes (G4s) are four-stranded DNA secondary structures, which are involved in a diverse range of biological processes. Although the anti-cancer potential of G4s in oncogene promoters has been thoroughly investigated, the functions of promoter G4s in non-cancer-related genes are not well understood. We have explored the possible regulatory roles of promoter G4s in cardiac function-related genes using both computational and a wide range of experimental approaches. According to our bioinformatics results, it was found that potential G4-forming sequences are particularly enriched in the transcription regulatory regions (TRRs) of cardiac function-related genes. Subsequently, the promoter of human cardiac troponin I (TnIc) was chosen as a model, and G4s found in this region were subjected to biophysical characterisations. The chromosome 19 specific minisatellite G4 sequence (MNSG4) and near transcription start site (TSS) G4 sequence (−80 G4) adopt anti-parallel and parallel structures respectively in 100 mM KCl, with stabilities comparable to those of oncogene G4s. It was also found that TnIc G4s act cooperatively as enhancers in gene expression regulation in HEK293 cells, when stabilised by a synthetic G4-binding ligand. This study provides the first evidence of the biological significance of promoter G4s in cardiac function-related genes. The feasibility of using a single ligand to target multiple G4s in a particular gene has also been discussed.

, J Chem Biol, Vol: 4, Pages: 159-165UNLABELLED: Inhibition of protein kinases in the fight against disease remains a constant challenge for medicinal chemists, who have screened multitudes of predominantly planar organic scaffolds, natural and synthetic, to identify potent-albeit not always selective-kinase inhibitors. Herein, in an effort to investigate the potential biological utility of metal-based compounds as inhibitors against the cancer-relevant targets mitogen-activated protein kinase and cyclin-dependent kinase 2, we explore various parameters in planar platinum(II) complexes with substituted phenanthroline ligands and aliphatic diamine chelate co-ligands, to identify combinations that yield promising inhibitory activity. The individual ligands' steric requirements as well as their pattern of hydrogen bond donors/acceptors appear to alter inhibitory potency when modulated. Zinc(ii) coordination polymers with pseudopeptidic ligands
, CrystEngComm, Vol: 13, Pages: 6997-7008Two new phenyl-bridged pseudopeptidic ligands have been prepared and structurally characterised. The nature of the ligands' substituents play an important role in the nature of the solid state structure yielding either hydrogen bonded linked sheets of molecules or infinite hydrogen bonded networks. Both these ligands were reacted with a range of zinc(ii) salts with the aim of synthesising coordination polymers and networks and exploring the role that anions could play in determining the final structure. The crystal structures of four of these systems (with ZnSO4 and ZnBr2) were determined; in one case, a 3D coordination network was obtained where zinc-ligand coordination bonds generated the 3D arrangements. Three other 3D networks were obtained by anion-mediated hydrogen bonding of coordination 1D chains or 2D sheets. These four very different structures highlight the important role played by the ligands' substituents and the counteranions present in the system.
