Most chronic diseases have been demonstrated to have a link to nutrition. Within food and nutritional research there is a major driver to understand the relationship between diet and disease in order to improve health of individuals. However, the lack of accurate dietary intake assessment in free-living populations, makes accurate estimation of how diet is associated with disease risk difficult. Thus, there is a pressing need to find solutions to the inaccuracy of dietary reporting.Metabolic profiling of urine or plasma can provide an unbiased approach to characterizing dietary intake and various high throughput analytical platforms have been used in order to implement targeted and non-targeted assays in nutritional clinical trials and nutritional epidemiology studies. This review describes firstly the challenges presented in interpreting the relationship between diet and health within individual and epidemiological frameworks. Secondly we aim to explore how metabonomics can benefict different types of nutritional studies and discuss the critical importance of selecting appropriate analytical techniques in these studies. Thirdly we propose a strategy capable of providing accurate assessment of food intake within an epidemioligical framework in order establish accurate associations between diet and health.

Bell J, Frost G, Thomas E, Brody L, Arisoylu M, Miller Aet al., 2012, Liposome encapsulated acetate (Lita) for the treatment of metabolic and oncogenic diseases

Cellular metabolic deregulation is a central premise in the development of many chronic diseases including cancer. Recently, there has been renewed interest in the potential of metabolic engineering, through the use of small molecules, to achieve metabolic retransformation of abnormal cells and tissues. However, making sufficient and sustain metabolic impact though this methods can be difficult, as it requires both identification of suitable small molecule(s) and an appropriate mode of delivery. Here we present a unique approach to tackle metabolic deregulation in vivo by the use of short-chain fatty acids (SCFAs) encapsulated in a liposomal nanoparticle delivery system. We demonstrate that liposomal delivery of the SCFA acetate leads to improved metabolic fitness of several organs, including liver and adipose tissue. Furthermore, treatment of mice consuming a high fat diet resulted in a significant reduction in hepatic fat accumulation, a reduction in inflammatory markers and modulation in key genes associated with fat metabolism, including UCP2, ACOX1 and CPT1A. These phenotypic and molecular events were underpinned by modulations in the epigenetic regulation of the liver. Finally, significant antitumor effects were observed in a xenograft murine model, including significant inhibition of tumour growth, following treatment. Our results demonstrate that this delivery system, liposomal encapsulated SCFAs, can be utilised as a novel method for the treatment of malignant transformation and metabolic deregulation in vivo.

Frost G, Wilkinson J, 2012, Compounds and their effects on appetite control and insulin sensitivity

Imperial innovations filed a UK priority patent application No1213629.7 entitled “compounds and their effects on appetite control and insulin sensitivity” in the name of Imperial Innovations and the University of Glasgow on the 31st of July 2012. We have filed a subsequent PCT application on the 31st of July 2013. The covers composition of matter and on a number of [novel prodrugs] or [propionate inulin esters] specifically designed to deliver short chain fatty acids to the gut. The patent also has claims directed to formulations and use in reduction of appetite/food intake/calorie intake. Claims are also directed and improving insulin sensitivity in a subject and for the treatment or prevention of diabetes or obesity.
