Summary

My PhD and postdoctoral work have focused on the study of barrier immunity, with a particular emphasis on the signals regulating the local establishment of protective T cell responses, in both infectious and inflammatory settings.

As a PhD student in the laboratories of Dr. Shabaana Khader and Dr. Jay Kolls at the University of Pittsburgh, my studies focused on the factors required for the induction of local protective immune responses against Mycobacterium tuberculosis (Mtb) and other pulmonary mycobacterial pathogens. I subsequently joined Dr. Sarah Gaffen’s lab at the University of Pittsburgh for a postdoc. There, I continued my interest in barrier immunity, focusing now on skin inflammation. I found that the endoribonuclease MCPIP1 is a regulator of IL-17A and IL-17F signalling in the skin.

In 2016, I moved to London, where I joined Professor Adrian Hayday’s lab for a postdoc. Specialising now on gd T cells, I described a population of IL-17-producing uterine gd T cells which play a non-redundant role in the protection of the female reproductive tract against fungal infection. More recently, I began to query whether there are general rules governing how gd T cells are targeted to specific tissues. By optimising gene transfer methods for the study of the T cell receptor, I demonstrated that conserved mechanisms underlie the interaction between gd T cells and the epithelium across different organs (manuscript under preparation).

Research interests

Our research focuses on gd T cells, which are disproportionately enriched at body surfaces. gd T cells emerge from the thymus precommitted to an effector fate and are independent from MHC for their activation. Taken together, these features make them attractive for cancer immunotherapy and indeed, several gd T cell-based products have entered clinical trials. We study how specific gd T cell populations are established in tissues, including the gut and the female reproductive tract, and the impact of the bidirectional convesations that they engage in with the epithelium. Understanding the mechanisms underlying the developmental associations of gd T cells with tissues could offer basic biological insights into tissue immunosurveillance and improved therapeutic approaches applicable to many anatomical sites.
