TheOMis tetheredto thepeptido-glycanviathelipoprotein,Lpp.However,theimportanceof theenvelope’smultilayeredarchitectureremainsunknown.Here,whenweremovedphysicalcouplingbetweentheOMandthepeptidoglycan,cellslosttheabilityto sensedefectsin envelopeintegrity.Furtherexperimentsrevealedthatthecriticalparameterforthetransmissionof stresssignalsfromtheenvelopeto thecytoplasm, wherecellularbehaviouris controlled,is theIM-to-OMdis-tance.Augmentingthisdistancebyincreasingthelengthof thelipoproteinLppdestroyedsignalling,whereassimultaneouslyincreasingthelengthof thestress-sensinglipoproteinRcsFrestoredsignalling.Ourresultsdemonstratethephysiological importanceof thesizeof theperiplasm.TheyalsorevealthatstrictcontrolovertheIM-to-OMdistanceis requiredforeffectiveenvelopesurveillanceandprotection,suggestingthatcellulararchitectureandthestructureof transenvelopeproteincomplexeshavebeenevolutionarilyco-optimisedforcorrectfunction.Similarstrategiesarelikelyat playin cellularcompartmentssurroundedby2 concentricmembranes,suchaschloroplastsandmitochondria.
